加拿大阿尔伯塔大学Todd Lowary课题组的最新研究报道了条件致病菌堪萨斯分枝杆菌合成十三糖脂低聚糖抗原。这一研究成果发表在2021年9月24日出版的国际学术期刊《德国应用化学》上。

课题组描述了从条件致病菌堪萨斯分枝杆菌中化学合成三糖LOS(2)的聚糖部分。目标包含许多非常见的结构主题,使它的组装复杂化。本报告描述了与堪萨斯州分枝杆菌相关的第一个合成,是迄今为止合成的最复杂的分枝杆菌LOS聚糖,考虑到独特单糖和糖苷键的大小和数量。

Title: Synthesis of a Tridecasaccharide Lipooligosaccharide Antigen from the Opportunistic Pathogen Mycobacterium kansasii

Abstract: The outer surfaces of mycobacteria, including the organism that causes tuberculosis, are decorated with an array of immunomodulatory glycans. Among these are lipooligosaccharides (LOS), a class of molecules for which the function remains poorly understood. We describe the chemical synthesis of the glycan portion of a tridecasaccharide LOS (2) from the opportunistic pathogen Mycobacterium kansasii. The target contains a number of unusual structural motifs that complicate its assembly. This report describes the first syntheses related to M. kansasii LOSs and 2 is the most complex mycobacterial LOS glycan to be synthesized to date when considering size and number of unique monosaccharides and glycosidic linkages. These studies not only provide a roadmap for the preparation of additional members of this family of glycans, but also provides a valuable probe for use in structure–activity relationship investigations
