癌细胞系百科全书数据库新增定量蛋白质组学数据

近日,美国哈佛医学院Steven P. Gygi、David P. Nusinow等研究人员合作新增了癌细胞系百科全书(CCLE)数据库中的定量蛋白质组学数据。该研究2020年1月23日发表于国际一流学术期刊《细胞》。

研究人员通过质谱法对来自不同谱系的375种细胞系中的数千种蛋白质进行定量分析,从而扩展了CCLE,并揭示了DNA和RNA方法未发现的信息。研究人员观察到RNA信息中大量缺失的通路内和通路之间的未知关联。对微卫星不稳定(MSI)细胞系的分析揭示了与突变和翻译监测相关特定蛋白质复合物的失调。这些以及其他蛋白质复合物与对几种不同基因敲低的敏感性有关。这些数据与更广泛的CCLE相结合,可作为探索细胞行为并促进癌症研究的广泛资源。

Abstract: Proteins are essential agents of biological processes. To date, large-scale profilingof cell line collections including the Cancer Cell Line Encyclopedia (CCLE) has focusedprimarily on genetic information whereas deep interrogation of the proteome has remainedout of reach. Here, we expand the CCLE through quantitative profiling of thousandsof proteins by mass spectrometry across 375 cell lines from diverse lineages to revealinformation undiscovered by DNA and RNA methods. We observe unexpected correlationswithin and between pathways that are largely absent from RNA. An analysis of microsatelliteinstable (MSI) cell lines reveals the dysregulation of specific protein complexesassociated with surveillance of mutation and translation. These and other proteincomplexes were associated with sensitivity to knockdown of several different genes.These data in conjunction with the wider CCLE are a broad resource to explore cellularbehavior and facilitate cancer research.
